General information

Storage conditions and duration: Store in a refrigerator at 2-8°C and away from light. This product can be used up to 14 weeks after production (if the sterile packaging is not opened).

Considerations for waste disposal: It should be done according to the laboratory waste disposal instructions.

Ingredients: This product contains bicarbonate buffer and HEPES, glucose, lactate and pyruvate needed to meet the nutritional needs of the egg in laboratory conditions outside the CO2 incubator.

The efficiency of the following protocol for the collection of cumulus-ovum masses (COCs) and use for their denudation has been demonstrated, and this protocol is presented only as an example:

1- Before use, the required amount of the culture medium should be mixed with the amount (volume-volume) of human serum albumin or HSA (Human Serum Albumin) with a concentration of 100 mg/ml to provide a protein supplement.
2- The required volumes of the culture medium should be dripped in culture dishes and then completely covered by mineral oil.
3- Before placing the masses of COCs, the dish prepared in the previous step should be placed in a CO2-free incubator for a sufficient period of time (1-2 hours) to ensure that the temperature balance is established. If using a CO2 incubator, make sure the lid of the container is completely closed in order to prevent pH drop.

Warnings


1- The user must read this manual completely and receive the necessary training before using the product.
2- This product cannot be used for injection into the body.
3- Do not re-sterilize the product.
4- Avoid using the product if you see the following:
- Presence of damage in sterile packaging
- The expiration date has passed
- Color change, turbidity or the presence of suspended particles or sediment.
5- This product contains the antibiotic gentamicin sulfate, so it should be used with caution in patients who are allergic to this antibiotic.
6- This culture medium has a small amount of sodium bicarbonate buffer and therefore, before use or during use, exposure to CO2 is not necessary, causing toxicity.
7- When using this product, in order to prevent contamination, aseptic methods should be used.
8- This product can be used safely up to 7 days after opening the sterile package.

Note: In most cases, the concentration of CO2 in the incubator at the rate of 5-7% will lead to the pH of the culture medium being in the accepted range of 7.2-7.4. However, fine-tuning the CO2 concentration to achieve an approximate pH of 7.3 (7.33 - 7.27) as the optimal pH depends on several factors, including the physical characteristics of the incubator and geographical altitude. As a result, it is recommended that each laboratory calculate the CO2 concentration required to achieve the optimal pH of 7.3 in the culture medium.
