Application:
Repronext kit is designed to isolate PRP for intrauterine autologous injection for infertility treatment applications. Platelet-rich plasma (PRP) is one of the most important blood derivatives. Platelets have more than 20 types of growth factors and other protein molecules, including cytokines and chemokines, which play a role in processes such as proliferation, differentiation and cell regeneration. The therapeutic ability of PRP depends on the amount of malignant factors in platelets, the most important of which are: platelet-derived growth factor (PDGF), transforming growth factor beta (TGFB), vascular endothelial factor (VEGF), insulin-like growth factor (IGF). 1) and hepatocyte growth factor (HGF).
The platelets in PRP begin to secrete growth factors with the addition of special activating factors such as thrombin and calcium chloride, which play a role in inducing and stimulating the release of growth factors from the granules in them. Over the past decade, the therapeutic potential of PRP and its resulting regenerative properties have been used in various fields of medical science, including infertility treatment, reconstructive and plastic surgery, oral and dental surgery, medical wool, hair transplantation, and orthopedics.

Storage conditions and duration: The kit components should be kept at room temperature.

The performance of the following protocol for the Repronext kit for isolation of PRP for autologous intrauterine injection is designed for infertility treatment applications and this protocol is provided as an example only:

1- First, the butterfly needle and the needle and the gadget should be connected to the gadget holder.
2- Disinfect the blood collection site with an alcohol pad from the center to the periphery.
3- The butterfly needle is slowly inserted into the vein and by connecting the tubes A to the holder and the nozzle, the blood sampling process is performed in the amount of 40 ml (avoid filling the tube completely).
4- Tubes containing blood and anti-coagulant should be gently corked several times.
5- Tubes containing blood should be centrifuged at g or RCF 150 for 10 minutes. (use a tube with an equal volume of water to create balance during centrifugation)
6- Gently remove the upper plasma and the coat tissue layer (white layer on the layer of red blood cells) using an angioket and ml10 syringe (to facilitate the process of plasma removal, use a syringe tip to remove the negative pressure inside the tube).
7- Transfer the plasma to tubes B and centrifuge at g or RCF 1200 for 15 minutes (at the end of this stage, a white deposit can be seen on the bottom of the tube).
8- Collecting the supernatant plasma using an angioket so that 1.5 ml of plasma remains on the white sediment.
9- Very slow swirling of the tubes to separate and homogenize the white precipitate in 1.5 ml of the remaining plasma (high speed during this step leads to the lysis of platelets, so usually the time required for this step It is about 10-15 minutes).
10- Collecting the required volume of PRP using insulin syringes for injection.

Warnings
