General information

Application: human embryo biopsy

Storage conditions and duration: Store in a refrigerator at 2-8°C and away from light. This product can be used up to 10 weeks after production (if the sterile packaging is not opened).

Considerations for waste disposal: It should be done according to the laboratory waste disposal instructions.

Ingredients: This product contains bicarbonate buffer and HEPES, human serum albumin (HSA), glucose, lactate and pyruvate and amino acids to meet the needs of the human embryo in laboratory conditions during the biopsy process.

The following protocol has been demonstrated to be effective for human embryo biopsy, and this protocol is provided as an example only:

1- Volumes of 25-50 µl of the fetal biopsy medium are dropped in the biopsy dish and then completely covered by mineral oil.
2- Before placing the embryos, the dish prepared in the previous step should be placed in the CO2-free incubator for a sufficient period of time to ensure the temperature balance. In case of using a CO2 incubator, make sure that the lid of the container containing the medium is completely closed in order to prevent pH drop.
3- The embryos are placed in the drop of biopsy medium and the biopsy process is performed after loosening the cell connections.
4- After the biopsy process is completed, the embryos are washed and kept in the incubator after being transferred to a fresh culture medium.

Warnings

1- The user must read this manual completely and receive the necessary training before using the product.
2- This product cannot be used for injection into the body.
3- Do not re-sterilize the product.
4- Avoid using the product if you see the following:
- Presence of damage in sterile packaging
- The expiration date has passed
- Color change, turbidity or the presence of suspended particles or sediment.
5- This product contains human serum albumin (HSA).
6- This medium contains small amounts of sodium bicarbonate buffer and therefore does not need to be exposed to CO2 to adjust the pH before or during use. Because this environment contains HEPES buffer, its exposure to CO2 causes toxicity.
7- This product contains the antibiotic gentamycin sulfate, so it should be used with caution in patients who are allergic to this antibiotic.
8- When using this product, in order to prevent contamination, aseptic methods should be used.
9- This product can be used safely up to 7 days after opening the sterile packaging.

Note: In most cases, the concentration of CO2 in the incubator at the rate of 5-7% will lead to the pH of the culture medium being in the accepted range of 7.2-7.4. However, fine-tuning the CO2 concentration to achieve an approximate pH of 7.3 (7.33 - 7.27) as the optimal pH depends on several factors, including the physical characteristics of the incubator and geographical altitude. As a result, it is recommended that each laboratory calculate the CO2 concentration required to achieve the optimal pH of 7.3 in the culture medium.
